The 0.25% alcohol-acid differentiation solution is a 70% alcoholic solution containing 0.25% concentrated hydrochloric acid. It is commonly used in histology and cytology to remove excess stain and highlight nuclei after hematoxylin staining in the H&E and PAP methods. To achieve H&E staining with precise nuclear details, it is crucial that certain hematoxylins are properly differentiated, making the alcohol-acid solution particularly effective.

Available in various concentrations of hydrochloric acid (0.25%, 0.5%, and 1%) in 70% alcohol.
