The 1% alcohol-acid differentiation solution is a 70% alcoholic solution containing 1% concentrated hydrochloric acid. It is commonly used in histology and cytology to remove excess stain and highlight nuclei after hematoxylin staining in the H&E and PAP methods. To achieve H&E staining with precise nuclear details, it is crucial that certain hematoxylins are properly differentiated, making the alcohol-acid solution particularly effective.
